Technically, what your employer did is illegal: the law probibits harassing (which can include mocking or making fun of) or discriminating against people with disabilities. Practically, if this a one time slip and he also apologized, even clumsily, for it, you would get very little, if any compensation for it, because compensation is related to harm, and the harm from one inappropriate joke is essentially none; therefore, it is not worth taking action over. Do keep notes and his email as evidence and for future reference; if the behaviour reoccurs and becomes a pattern of harassment, or you suffer some negative employment consequences, like being passed over for a promotion, you may then have a worthwhile claim.
